Topoisomerase Definition:

  • Untwisting of the double helix causes tighter twisting and strain ahead of the replication fork. Topoisomerase helps relieve this strain by breaking, swiveling, and rejoining DNA strands.

Topoisomerase Notes:

Many important enzymes participate in DNA replication including topoisomerases. These are the enzymes that participate in the over winding or underwinding of DNA. The winding problem of DNA arises due to the intertwined nature of its double-helical structure and it may get coiled or overwound. During DNA replication and transcription, DNA becomes overwound ahead of a replication fork and this is where topoisomerases come to play and help in easing the coils of DNA to prevent DNA damage.
