Affinity labeling is a strategy to chemically modify an amino acid residue, within a ligand-binding site in an enzyme. This can happen either at a regulatory allosteric site or at the active site. The process involves a reagent that structurally resembles the natural ligand of the enzyme, but at the same time features a functional group that is capable of covalently reacting with many different amino acid residues.
