Amber Suppressor is a mutant allele, coding for a tRNA, bearing an anticodon to recognize the amber codon (UAG), and eventually suppress amber mutations. The anticodon is altered, such that an amino acid is inserted at an amber codon in translation, by the suppressor tRNA,, preventing termination.
