Chain Termination DNA Sequencing Method Definition:

  • A DNA-sequencing method that uses dideoxynucleotides to cause termination of DNA replication at random sites.

Chain Termination DNA Sequencing Method Notes:

Chain Termination method, widely known as the Sanger Sequencing is a method of DNA sequencing, based on the incorporation of selected chain-terminating dideoxynucleotides by DNA polymerase during in vitro DNA replication. This method was developed by Frederick Sanger and colleagues in 1977 and first commercialized by Applied Bio systems. It was the most used method used for sequencing for approximately 40 years. Recently this method has been replaced by "Next-Gen" sequencing methods for large scale, automated analyses. Meanwhile, the Chain Termination method is still used for smaller scale projects and for validation of Next-Gen results.
